Systematic study of the physicochemical properties of a homologous series of aminobisphosphonates

ABSTRACT: Aminobisphosphonates, e.g., alendronate and neridronate, are a well known class of molecules used as drugs for various bone diseases. Although these molecules have been available for decades, a detailed understanding of their most important physicochemical properties under comparable conditions is lacking. In this study, ten aminobisphosphonates, H2N(CH2)nC(OH)[P(O)(OH)2]2, in which n = 2-5, 7-11 and 15 have been synthesized. Their aqueous solubility as a function of temperature and pH, pKa-values, thermal stability, IR absorptions, and NMR spectral data for both liquid (1H, 13C, 31P-NMR) and solid state (13C, 15N and 31P-CPMAS NMR) were determined. Poor aqueous solubility (<0.1 mg/mL) affects a significant number of drugs currently on the market or under development. Several formulation strategies including salt formation, particle size reduction, and solid dispersion approaches have been employed with varied success. In this review, we focus primarily on the emerging trends in the generation of amorphous and micro/nano-crystalline solid dispersions using electrospinning to improve the dissolution rate and in turn the bioavailability of poorly water-soluble drugs. Molecular surface calculations on organic compounds

Abstract The molecular surface area was calculated for 82 hydrocarbons, esters, ethers, alcohols and ketones including linear, branched and cyclic ones. The relationship between the aqueous solubility and the molecular surface area was determined for each family of compounds and for all the families together. The results show that solubility is mainly determined by molecular surface area and that the influence of functional group is the same for all the oxygen containing compounds studied.
